Cas9-Cell line Gene Mutation service（CGM）基因点突变细胞系是细胞基因研究方法中非常有用的一个手段，可以在细胞内原位对点突变的功能进行研究。在细胞系上实现高效的转染是实现基因点突变的前提；提高效率要解决问题是：Cas9切割位点与“点突变”位点之间距离导致点“点突变”效率大幅下降的问题，还有“点突变”载体的选择问题。
目前使用Cas9的引入基因“点突变”的具体实验实践中的“单碱基突变”的效率仍然受到很多要素的影响，例如是否在点突变附近是否存在有合适的gRNA切割位点、切割位点和突变位置之间距离、不同细胞系之间的Cas9切割活性效率差异的问题、还有细胞株是否存在“假基因”和基因“多拷贝”的问题导致无法获得纯合的问题等等。
Cas9X针对不同单碱基突变的情况使用不同的Mutation载体策略：包括短同源臂可以使用Oligo引入突变，需要长同源臂的使用ssDNA引入突变。而针对难以操作的细胞系，将需要通过先构建Cas9稳转细胞株再进行gRNA和载体共转染来提高效率。Cas9X针对每个具体的位点、每个具体的细胞优化实验条件用来满足不同细胞系和不同位点突变的服务需求。

项目名称：构建JAK2基因点突变(V617F)的人成巨核细胞白血病细胞MEG-01
· 设计与转染：设计了gRNA和ssDNA供体模板，以靶向JAK2基因的特定区域并引入V617F突变（图1）。将它们共同电穿孔到MEG-01细胞中。
· 单细胞克隆：通过使用“克隆增强”技术分离出单个细胞，以确保细胞的单克隆性。
· 筛选与扩增：在96孔板中的单克隆细胞群通过Sanger测序进行筛选（图2）。经确认带有V617F突变的阳性克隆被扩增以进行进一步分析。
